Check if you have it enabled. If you don't know where, go to "Multiplayer" then "Customize", at the bottom left, it should say "Enable voice for this mod". It should have a check on the box.

I suggest you type «voice_loopback 1» in console if you want to test your mic in a game. You'll hear what the other players hear ( or do not hear ) when you press the button.

As silly as it sounds, check that your mic is plugged into the right spot. I had to disconnect my mic for some reason, and played a good five or six games where it seemed that I couldn't get any backup, not even while defending a hive, no matter how much I yelled into the mic. Only after a few hours did someone tell me that it wasn't just screwy mic settings; they could here nothing AT ALL. Turns out I plugged the mic into some other spot, instead of the right one <!--emo&:)--><img src='http://www.unknownworlds.com/forums/html/emoticons/smile.gif' border='0' style='vertical-align:middle' alt='smile.gif'><!--endemo-->

    Try this one,

a friend borrowed the mic, gave it back so i put it on the desk and put the cable down the back.
A few days later I tried to use it and nothing happend, someone suggested it wasent plugged in and I of course denied it then it clicked! Turns out most ppl in the clan had done the same thing before.

A friend of mine had the same issue, the mic worked in windows but not in HL. So as we use the same keys I simply sent him a copy of my config.cfg and it all worked. I think the relevent entries are below, if they aren't in then add them (obviously change the bind key to your preference.

    voice_scale "2.000000"
    voice_enable "1"
    voice_forcemicrecord "1.000000"
    voice_modenable "1"
    bind "g" "+voicerecord"
